What are the production methods of lipase

Apr 11, 2024

1. Microbial fermentation method: This is the main method for producing lipase in industry. Lipase is produced by selecting appropriate microbial strains, such as Yarrowia lipolytica, Pseudomonas, etc., and through a fermentation process. Microbial fermentation method has the advantages of short production cycle, high efficiency and low cost.
2. Animal and plant extraction method: Although lipase is also contained in animals and plants, this method is not commonly used in industrial production due to the high extraction cost and seasonal and regional restrictions.
3. Genetic engineering method: Through genetic engineering technology, the lipase gene is cloned into a suitable expression vector, and then expressed in host cells (such as Escherichia coli, Saccharomyces cerevisiae, Pichia pastoris, etc.). This method can achieve efficient production of lipase, but may face challenges in expression volume and enzyme activity.
4. Immobilized cell method: Using immobilization technology, lipase-producing microbial cells are fixed on a certain carrier for repeated use. This method can improve the stability and reusability of lipase.
5. Extraction and purification of lipase: Extract lipase from fermentation broth or animal and plant tissues, and purify it through a series of steps such as centrifugation, filtration, chromatography, etc., to obtain high-purity lipase products.
6. Improvement of lipase stability: Improve the thermal stability and organic solvent tolerance of lipase through protein engineering or chemical modification to adapt to different industrial application needs.
7. Lipase surface display technology: fix lipase on the cell surface to improve its stability and substrate conversion rate in industrial applications.
8. Whole-cell catalysis of lipase: Using whole cells containing lipase as a catalyst, this method has the advantages of simple preparation and low cost.
